Cytotoxic activity of seed extracts of Polyalthia longifolia

The purpose of the study is to evaluate the cytotoxic activity of seed extracts of Polyalthia longifolia . Different seed extracts (petroleum ether, chloroform,methanol and aqueous) of P.longifolia were prepared and evaluated for cytotoxic activity. The activity was investigated on Vero, HEp2 and HeLa cell lines by MTTassay and DNA fragmentation analysis. The MTT assay showed that methanol extract was less toxic towards the normal (Vero) cell lines while the others moretoxic. Hence the methanol extract was tested on HEp2 and HeLa cell lines and showed the IC50 values of 0.3125 mg/ml and 0.625 mg/ml respectively. The DNAfragmentation analysis showed that there was a necrotic as well as apoptotic cell death when the methanol extract was evaluated on HEp2 and HeLa cell lines. Thestudy confirms the cytotoxic activity of different seed extracts of P.longifolia and can be used for further drug formulation studies.
