Effect of superdisintegrants in the formulation of taste-masked orodispersible tablets of Tizanidine HCl

The objective of this study was to prepare orodispersible tablets (ODTs) which can rapidly disintegrate in saliva using taste-masked granules of drug, Tizanidine HCl (TZN HCl). The taste-masked granules were prepared using gastro-erodible aminoalkyl methacrylate copolymer (Eudragit E-100) in different ratios by extrusion method. Drug-polymer complexes (DPCs) were tested for drug content & in vitro drug release in simulated salivary fluid (SSF) of pH 6.8. Complex that did not release drug in SSF was considered taste-masked and selected for formulation of ODTs. The effect of various superdisintegrants such as Chitosan, Croscarmellose sodium (CCS) & Sodium starch glycolate (SSG) in formulation of orodispersible tablets was also studied. The prepared tablets were evaluated for weight variation, thickness, hardness, friability, wetting time, disintegration time, water absorption ratio, and in-vitro drug release. Among all, the formulation F9 containing (8% w/w concentration of chitosan) was considered to be the best formulation, with disintegration time of 22 seconds and in-vitro drug release of 99.97% in 10minutes in simulated gastric fluid (SGF).Thus, results conclusively demonstrated successful masking of taste & rapid disintegration of the formulated tablets.
