Formulation and evaluation of desloratadine orodispersible tablets by using ß-cyclodextrin and superdisintegrants

The aims of this study are (1) taste masking of highly bitter drug Desloratadine by ß-Cyclodextrin and (2) to prepare Orodispersible tablets of Drug: ß-Cyclodextrin complex. Desloratadine was complexed with ß-Cyclodextrin by kneading method. Drug: ß-Cyclodextrin in the ratio of 1:8 w/w was selected forfurther studies. The complex was evaluated for bulk density, angle of repose, drug content and in-vitro drug release at gastric pH (pH 1.2) and characterized byInfrared spectroscopy and Differential Scanning Calorimetry. The tablets contained Desloratadine: ß-Cyclodextrin complex and one of the three superdisintegrants(Ac-Di-Sol, Polyplasdone XL and Primojel) and other excipients. Tablets were compressed on a multistation rotary press. The tablets were evaluated for weightvariation, hardness, friability, thickness, disintegration time and dissolution at gastric pH. Desloratadine: ß-Cyclodextrin complex showed 100% of drug releasein 4 minutes in gastric pH. Tablets containing of Ac-Di-Sol (9%) disintegrated in 33 seconds and released 100% of drug in 5 minutes at gastric pH. Orodispersibletablets can be prepared with Desloratadine: ß-Cyclodextrin complex by using Ac-Di-Sol as disintegrant.
