ABSTRACT
Tannase is used in leather, food, beverages, pharmaceutical and chemical industry. It also helps in digestion and acts as antimicrobial agent. For its large Scale production and its best optimized production conditions was done for the Bacterial Tannase under submerged fermentation .Production was carried out in Tannic acid media which was the most appropriate media and it supported the highest growth of desired bacteria in presence of nutrient agar. Carbon source was glucose as Glucose was easily taken by bacteria; they showed faster growth at the incubation temp of 35 o C with the pH 4 and 5. Purification was done with ammonium sulfate fractionation and ammonium salts were removed by Dialysis. Ion Exchange chromatography was performed to justify the presence of proteins and enzyme. The molecular weight of tannase was identified by running the gel with standard molecular marker in polyacrylamide gel electrophoresis.
