Preparation and Characterization of Chitosan Microspheres Crossed-linked with 2, 3 - Dihydroxypropanal as a competitive Drug Delivery System

Chitosan microspheres with small particle size and good sphericity were prepared by a spray-drying method followed by treating with a cross-linker agent. To overcome the restriction in usingcrosslinked chitosan microspheres in the pharmaceutical field, 2,3-Dihydroxypropanal , a biocompatible reagent was used. The parameters investigated affecting crosslinking extents were thecrosslinking time and the concentration of the crosslinker agent. Glutaraldehyde, the most frequently aldehyde employed as chemical crosslinker agent for proteins, was also used as a control.The crosslinked spray-dried chitosan microspheres were analysed with respect to their morphological aspects, particle size, zeta potential and water uptake capacity. It was found that either the increase of the 2,3-Dihydroxypropanal concentration and the crosslinking duration decreased both the swelling capacity and the zeta potential of the chitosan microspheres.Compared to glutaraldehyde, 2,3-Dihydroxypropanal appears to be a good crosslinker reagent for chitosan microspheres with the advantage of being non-toxic.
