ABSTRACT
The neurodevelopmental hypothesis of schizophreniaproposes that interactions between genetic andenvironmental events occurring during the critical earlyperiod of brain development may negatively influenceneuronal growth, lamination and orientation, resultingin cytoarchitectural defects. The Wnt signalling pathwayis essential for normal embryonic development of thecentral nervous system. Disturbances of Wnt signallinghave been thought to be involved in producing thecytoarchitectural defects observed in schizophrenia. Thisarticle reviews current data on the alteration in the Wnttransduction cascade, which may represent an aberrantneurodevelopment in schizophrenia. Firstly, thecanonical Wnt signalling cascade is described. Secondly,recently published studies on the potential role of theWnt pathway in schizophrenia pathology are reviewed.Finally, the results of the latest experimental studies onthe influence of antipsychotics on the main protein ofthe Wnt pathway are outlined.
