Keywords: Vitex negundo Linn. | DPPH assay | total antioxidant activity | FRAP value | total phenolics
ABSTRACT
The methanolic extract of Vitex negundo L. was dissolved in distilled water and partitioned with n-hexane, chloroform, ethyl acetate and n-butanol successively. The antioxidant potential of all these fractions and remaining aqueous fraction was evaluated by four methods: 1,1-Diphenyl-2-picrylhydrazyl radical (DPPH) scavenging activity, total antioxidant activity, ferric reducing antioxidant power (FRAP) assay and ferric thiocyanate assay along with the determination of total phenolics. The results revealed that ethyl acetate soluble fraction showed highest percent scavenging of DPPH (86.0 ± 0.33%) at the concentration of 30 µg/ml. The IC50 value of this fraction was (15.54 ± 0.10 µg/ml) relative to butylated hydroxytoluene (BHT), having IC50 of 12.1 ± 0.92 µg/mL. Chloroform soluble fraction was found to have the highest total antioxidant activity (2.071 ± 0.13). Ethyl acetate soluble fraction also exhibited highest FRAP value (3367.8 ± 5.89 μg of trolox equivalents), highest concentration of total phenolic compounds (146.8 ± 2.09 mg/g) as well as highest percentage of inhibition of lipid peroxidation (58.37 ± 0.12%) as compared to other fractions.
