ABSTRACT
For the production of Polyhydroxybutyrate (PHB), different microorganisms were isolated from garden soil. After the morphological and biochemical studies, isolates were identified as Bacillus cereus, Bacillus megaterium, Ralstonia eutropha, Rhizobium spp. and P1. Organism P1 was isolated from the chemically contaminated water collected from industrial area, Hazira, Surat. These species were further explored for their potential to produce PHB. Extraction of PHB was done by hot chloroform method and was further quantified using crotonic acid assay. Highest cellular content was obtained from Ralstonia eutropha. Homology modeling of R. eutropha was done using various bioinformatics tools. The enzyme Acetyl CoAacetyltransferase encoded by gene phbAwas responsible for PHB production in R. eutropha. 3-D structure of protein was predicted by SWISS MODELand visualized by RASMOLtool. Ligand binding sites in particular enzyme were found out by using Qsite finder and protein was verified by Ramachandran plot
