It is well established that the role of psychosocial factorsis one of the most crucial compounds in the complexetiopathogenesis of depression. The aim of this article isto review data concerning the influence of stress andenvironmental and genetic factors on the appearance ofdepressive symptoms. This article describes the role ofboth: stressful life events end early stressful life eventssuch as sexual abuse, physical and psychological abuse,or education neglect in childhood. Later in the paper, therole of the hypothalamic-pituitary-adrenal axis is describedon account of its close relationship with mechanisms ofstress. Emerging evidence suggests that changes in thePPN axis constitute a mechanism mediating betweenstressful life events and increased vulnerability to stressand biological changes observed in depression. Theauthors also underline that psychosocial factors seem topredispose to illness only in combination with theinherited genetic predisposition to depression; thereforethe authors of this article paid special attention to the roleof genetic factors in response to early childhood stress andlife factors appearing in adult life. The described reviewconfirms the essential role of interaction of polymorphismsof 5-HTT, COMT, MAOA, CRHR1 genes andenvironmental factors in development of depressivesymptoms, and this indicates the need for further studiestaking into consideration interactions of these genes withstressing factors.
